化岩胶囊对骨肉瘤细胞系诱导调亡作用的实验研究

WITHERING-INDUCING EFFECT OF "HUA YAN JIAO NANG" ON THE CELL LINE OF OSTEOSARCOMA: An Experimental Study

为研究化岩胶囊对成骨肉瘤细胞系U2 OS的诱导凋亡作用。将不同浓度的化岩胶囊作用于成骨肉瘤细胞系U2 OS,应用形态学观察、台盼蓝染色、流式细胞术、原位末端标记 (TUNEL)等方法 ,检测其对骨肉瘤细胞的诱导凋亡的时间效应和剂量效应。结果显示化岩胶囊对U2 OS细胞有明显时间依赖和剂量依赖的生产抑制及诱导凋亡作用 ,细胞周期分析出现G2 /M期阻滞、明显的凋亡峰 ,形态学表现细胞核碎裂、染色体凝聚的典型凋亡特征 ,并出现大量多核细胞。免疫组织化学检测化岩胶囊诱导后细胞核中有广泛DNA断裂。表明化岩胶囊对骨肉瘤细胞有明显促凋亡作用。其作用表现出时间依赖和剂量依赖关系。

In order to study the withering-inducing effect 'HUA YAN JIAO NANG' (HYJN, Capsules for Eliminating Bone Tumors) on the cell line of osteosarcoma (U2-OS), different concentrations of HYJN were added to the culture fluid to act on the cultivated U2-OS, and the time-effect and dose-effect of HYJN inducing the withering of U2-OS were detected with morphological observations, trypan-blue staining, Flow cytometry (FCM), in-situ nick end labeling or TUNEL, etc. The results showed that HYJN had a significant production-inhibiting and withering-inducing effect of HYJN on U2-OS, which was obviously dependant on the time and dose; the G2/M-staged block and remarkable withering peak appeared in the cell cycle analysis; the morphological findings included some characteristics typical to the withering, such as broken cellular nuclei, chromosome aggregation, etc, and the appearance of lots of multinuclear cells; and the immunohistochemical detection showed extensive DNA rupture in the cellular nucleus after HYJN induction, suggesting that HYJN has a significant withering-stimulating effect on osteosarcoma cells, which was dependant on the time and dose..
