组织培养中TA对细胞脱分化、分化及生长的生理效应

摘要 试验表明,在枫叶海棠(Begonia heracleifia)和中华猕猴桃(Actinidia chinensis planch)的组织培养中三十烷醇(代号TA)单独使用及与其它激素配合使用时对细胞脱分化、分化及生长的生理效应如下:(1)TA单独使用时,茎段组织细胞无脱分化诱导作用,但与BA、Zt、NAA等激素配合使用时则表现出很强的协同增益效应,协同促进组织细胞的脱分化、分裂和生长。(2)TA单独使用时,对中华弥猴桃愈伤组织分化茅无诱导作用,但与ZL配合使用则具有促进茅分化和小苗生长的协同作用,还能使试管苗叶片较肥厚浓绿、健壮,对移栽成活有利。(3)TA单独使用对中华猕猴桃试管苗发根的诱导作用次于NAA,但与NAA配合使用却有显著的协同增益效应,发根时间缩短、发根的数量增多。(4)TA具有显著的促进根伸长的作用,与NAA配合使用则效果更住。 physiological effect of cell differentiation and dedifferentiation of Begonia heracleifolia and Actinidia chinensis in tissue culture with the present of TA alone, or and other plant growth hormons; (1)when only TA was provided to the culture medium neither of the two plant could be induced to dcdifferentiation, but when TA was applied together with 6-BA, ZT, NAA it showed a strong affect on inducing dediffercntiation of stem tissues and co-accelrating cell devision and growth. (2) TA alone could not induce callus differentiation of A. chinensis. It could, however, enhance the differentiation when cooperated with ZT. The number of seedling differentiation from the callus, the height of the seedling and leave number were all possitively correlated to TA conccntratio, which clearly indicate the co-enhancement of TA with ZT in accelerating bud differentiation and seedling growth, The leaves of seedling in TA tube were deep green, strong and thick which certainly increase the survival rate of the transplanted seedling. (3) TA was found less affectively than NAA in root development of A. chinensis. The effect was greatly increased when it was used together with NAA. It is indicated that: (1) when TA and NAA were applied at the same time, days for the development of root was greatly shorter. (2) TA alone has hardly any effect on the number of roots developed. It is better when TA was used with NAA than either TA or NAA alone, (3)TA could effectively accelerate the growth of roots and the effect would be even better when it was used with NAA.
