采用细胞融破酶底物法测定小鼠淋巴细胞白血病(P_(388))细胞、人粒细胞白血病(K_(562))细胞和相应的正常组织细胞—小鼠脾脏单个核细胞(MSMN)和成人末梢静脉血多形核细胞(ABPN)的Na^+,K^+-ATP酶活性,结果表明P_(388)和K_(562)细胞的Na^+,K^+-ATP酶活性明显高于MSMN和ABPN。采用^(86)Rb示踪法测定4种体外培养的肿瘤细胞—人肝癌细胞(SMMC-7721)、人胃腺癌细胞(SGC-7901)、人宫颈癌细胞(HeLa cells)和K_(582)细胞的 Na^+,K^+-ATP酶活性,并同它们的生长速度进行比较,发现肿瘤细胞生长速度较快者(SMMC-7721),Na^+,K^+-ATP酶的活性也较高。本文结果表明,肿瘤细胞具有高Na^+,K^+-ATP酶的特征,且同生长速度有关。
The activities of Na^+, K^+-ATPase of mouse P_(388) lymphocytic leukemia cells (P_(388) cells), human K_(562) granulocytic leukemia cells (K_(562) cells) and corresponding normal tissue cells——mouse spleen mononuclear cells (MSMN) and adult peripheral blood polymorphonuclear cells (ABPN) were deter- mined by using enzymatic substrate method in disrupted cells in this article. The results showed that the activities of Na^+, K^+-ATPase of P_(咐88) and K_(562) cells were significantly higher than those of MSMN and ABPN. The activities of Na^+, K^+-ATPase of four kinds of human tumor cells cultured in vitro--hepatic carcinoma cell line (SMMC-7721), gastric adenocarcinoma cell line (SGC-7901), cervical carcinoma cell line (HeLa cells) and Ks6: cells were examined by using 'Srubidium tracing method, and corralated with their growing speed. It was discovered that the faster growing speed of tumor cells (SMMC- 7721), the higher activity of Na^+, K^+-ATPase. Our findings suggested that these tumor cells had a significantly high activity of Na^+, K^+-ATPase which related to growing speed of tumor cells.
