摘要
以宁夏农科院选育的新品种宁杞 1号 3年生树为试材 ,对影响枸杞胚细胞悬浮培养及其再生系统建立的培养基与激素配比进行了研究。结果表明 ,以人工授粉后 18d的幼果 ,剖取成熟胚作为外植体 ,选择 MS+6 BA1.0 mg/ L+IBA0 .5 m g/ L 培养基诱导愈伤组织 ,经单细胞悬浮培养建立了稳定的细胞系 ;MS+6 BA0 .2mg/ L 培养基较为适宜枸杞胚状体萌发成苗 ,该再生苗转移到 MS+NAA0 .2 mg/ L 生根培养基上 ,诱导生根并获得完整植株 ;共移栽成活 14株 ,成活率达 38.8%。
The three year old trees of Lycium barbarum L.cv.Ningqi No.1 were used as plant materials.The MS mediums with different hormone composition were studied to compare their effect on establishment of the suspension culture on cell line from embryo in Lycium babarum L.The results showed that the 18 day old mature embryos from the fruit pollinated were taken out as explants,and inoculated in the best MS+6BA 1.0 mg/L+IBA 0.5 mg/L medium for inducing callus,the stable cell lines by the cell suspension culture were set up,MS+6BA 0.2 mg/L was the suitable medium for produced embryoids germination and seedling,development,MS+6BA 0.2 mg/L as rooting medium was used to culture the formed seedlings and 14 plantlets transferred to the field were obtained,the regeneration efficiency was 38.8%.
