以神经毒素 6_OHDA诱导PC12细胞凋亡长期被用作帕金森氏病 (PD)研究模型。由于氧应激在PD发病中起重要作用 ,在 6_OHDA处理前提前 3 0min加入不同剂量的保护剂EGCG ,对 6_OHDA造成的PC12细胞损伤有一定的保护作用 ;流式细胞术检测细胞凋亡率 ,结果表明EGCG表现出剂量依赖的保护作用 ,且在 2 0 0 - 4 0 0 μM时保护作用最佳 ;
The biological activities of (-)_Epigallocatechins gallate (EGCG) were investigated by observing the range of 6_OHDA_induced apoptosis of PC12 cells in the Parkinson's disease in vitro model. Measured by MTT assay, 6_OHDA inhibited cell viability in time_ and concentration_dependent manner. When PC12 cells were pretreated with EGCG for 30min before exposured to 250 μM 6_OHDA, results showed that EGCG had obvious concentration_dependent protective effects in the range of 50-400 μM; 200-400 μM EGCG showed best effects. In addition, typical apoptotic characteristics of PC12 cells were determined by fluoresence microscopy, after the cells treated with 250 μM 6_OHDA for 24 h or pretreated with EGCG before it. Results showed preincubation with 200-400μM EGCG led to significant inhibitory effects against PC12 cell apoptosis.
