提取猕猴桃体细胞杂种叶片总 DNA,PCR和限制性内切酶 ,分析了叶绿体基因组的trn T(UGU)和 5’trn L(UAA)外显子之间的 a~ b间隔区 DNA片段和光合系统 D1蛋白基因(psb A)片段 ,以及线粒体基因组的 ORF2 5片段的遗传特征 .结果表明 ,狗枣猕猴桃 (A.kolomikta)与中华猕猴桃 (Actinidia chinensis)的对称体细胞杂种具有与中华猕猴桃相同的 a~ b间隔区 DNA片段 ,叶绿体 DNA遗传为非随机分离 .还讨论了 cp
Total DNA is extracted from the leaves of Kiwi interspecific somatic hybrids between A. chinensis and A. kolomikta or A. chinensis and A. deliciosa . Polymerase chain reactions and its products of restriction fragment length polymorphisms are made for three DNA fragments in chloroplast and mitochondria genomes. The results show that as for the spacers between trnT(UGU)and 5′trnL(UAA)exon (a^b), the genetics of cpDNA is nonrandomly separated among the symmetric somatic hybrids between A. chinensis and A. kolomikta because their PCR patterns are similar to that of A. chinensis . The relationship between cpDNA genetics and the origin of Kiwi species is also discussed.
