摘要
目的 探讨致痫剂马桑内酯引发体外培养小胶质细胞激活的细胞内机制。方法 应用马桑内酯 ( CL ,2 5μmol/ L )作用于培养的新生大鼠的小胶质细胞 ,免疫细胞化学观察小胶质细胞的形态学改变及其核转录因子 NF-κB的表达情况。结果 CL刺激后小胶质细胞形态多为阿米巴形活化表现 ;刺激 10 m in内 NF-κB核表达细胞显著增多 ( P<0 .0 1) ,1h达高峰 ( 97.3 7± 1.94) % ,PDTC ( 10 0μmol/ L )可抑制其核表达。结论 致痫剂马桑内酯可快速激活小胶质细胞 ,其激活的细胞内机制可能与核因子
Objective To explore the intracellular mechanism of activation of isolated microglial cells induced by Coriaria lectone (CL). Methods Cultured neonatal microglial cells were acted by CL (2.5×10 -5mol/L) and their morphological changes and nuclear factor (NF)-κB expression were observed by immunocytochemical technique. Results After CL stimulation the microglial cells appeared as activated amebiod shape. Within 10 min after stimulation NF-κB p65-ir nuclear positive cells were increased significantly (P<0.01) and reached peak value at l h (97.37±1.94)%. PDTC (100 μmol/L) could inhibit the NF-κB expression in nuclei. Conclusion CL might quickly activate microglial cells, the intracellular mechanism of which might be related to the activation of NF-κB.
