摘要
目的 为探讨人端粒酶 RNA( h TR)基因在男性不育症患者睾丸组织中的表达及其意义。方法 应用核酸原位杂交技术对 47例男性不育症患者睾丸组织和 10例正常睾丸组织中端粒酶 h TR基因的表达进行检测和定位 ,并应用 HPIAS- 10 0 0高清晰度图像处理系统对 h TR阳性信号进行图像分析。结果 端粒酶 h TR基因阳性表达与生殖细胞(精原细胞、精母细胞、精子细胞 )的分布定位一致 ,在精细胞不发育 (唯 Sertoli细胞综合征 )患者睾丸组织中无端粒酶 h TR基因的表达。结论 提示端粒酶活性的缺乏可能是生殖细胞发育停滞的原因 ,但不是唯一原因 ,可能还有端粒酶以外的因素起作用。对端粒酶研究有助于了解它在男性生殖中的作用 。
Objective To study the expression of telomerase gene hTR in testes of infertile male and its significance. Methods By using in situ hybridization (ISH) techniques, the expression of telomerase gene hTR mRNA in testes of 47 infertile males and 10 normal testicular tissues were detected. Results In male testes, there was a positive correlation between the expression of hTR and the quantity and density of germ cells (spermatogonia, spermatocyte, spermatid). The expression of hTR in some germinal cell maturation arrest patients was not significantly different from that in normal testes. Conclusion Deficiency of telomerase might be a factor but not the only factor for germinal cell maturation arrest. This study provides experimental groundwork for the gene therapy of male infertility.
