摘要
本文报道了从人工培养的灵芝菌丝体中经生理盐水浸取,硫酸铵盐析,DE-52离子交换层析,Sephadex G-100柱层析,分离出一种活性组分,称之为灵芝血凝蛋白(Ganoclerma Hemoagglutination protein简称GHAP),Sephadex G-100测得其分子量为126000,SDS-PAGE得两条色带,分子量分别为55600和59800,等电聚焦电泳测得等电点为4.6,糖含量为2.56%。GHAP对兔红细胞有很强的凝集作用,体外细胞培养发现它具有抑制腹水型肝癌细胞(AH)增长和促进淋巴细胞分裂的作用。
Ganoclerma hemoagglutination protein (GHAP) was obtained by extraction with normal saline, precipitation with ammonium sulfate, ion-exchange chromatography on DE-52, and gel filtration chromatography on SephadexG-100. Ployacrylamide electrophresis gave one band and sodium dodecyl sulfate ployacrylamide electrophresis gave two bands, with molecular weights of 55, 600 and 59, 800. SephadexG-100 gel filtration chromatography gave it' s molecular weight of 126, 000, the isoelectric point being about 4. 6. The GHAP agglutinated rabbit erythrocytes remarkably, inhibited the growth rate of AH cultured cell and promoted Lymphocyte division in vitro.
