尼尔雌醇皮埋剂保护去卵巢大鼠的骨量 被引量:1

摘要 目的 探讨尼尔雌醇 (NYL )皮埋剂对卵巢切除 (OVX)大鼠骨量的影响 ,为长期雌激素替代治疗提供一种新的选择。方法 用 DEX-A分别测定假手术 (Sham)组、卵巢切除 (OVX)组、卵巢切除加左炔诺孕酮皮埋剂 (LNG)组、卵巢切除加 L NG皮埋剂和 NYL皮埋剂 (LNG+NYL )组及 OVX加 NYL皮埋剂 (NYL)组大鼠的活体和离体骨密度 (BMD) ,并对血清碱性磷酸酶 (AL P)和尿吡啶啉 /肌酐 (Pyr/ Cr)进行检测。结果 与Sham组比较 ,OVX组和 LNG组大鼠全身、腰椎活体 BMD和腰椎、股骨整体及各兴趣区 (除第 3兴趣区 )、胫骨整体及干骺端第 1兴趣区离体 BMD均显著下降 (P<0 .0 5~ P<0 .0 1 ) ;而血清 ALP和尿 Pyr/ Cr增高 ,后者差异有显著性 (P<0 .0 5)。 L NG+NYL皮埋剂组和 NYL皮埋剂组 BMD较OVX组增加 ,在许多部位差异有显著性 (P<0 .0 5~ P<0 .0 1 ) ,BMD与 Sham组接近 ;而它们的血清 ALP和尿 Pyr/ Cr较 OVX组下降 (P<0 .0 5~ P<0 .0 1 )。结论  LNG皮埋剂不能阻止 OVX导致的骨丢失 ;NYL皮埋剂有保护 OVX大鼠骨量的作用。 Objective To investigate the effects of nylestroil implant on the bone loss of ovariectomized (OVX) rats and provide a new choice for estrogen replacement treatment of needed a longer time. Methods The dual energy X ray absorptiometry was adopted to measure bone mineral density (BMD) in vivo and vitro of Sham operated (Sham )group and OVX group and OVX treated with levonorgestrel impant (LNG )group and OVX treated with levonorgestrel implant and nylestroil implant (LNG+NYL) group as well as OVX treated with nylestroil implant (NYL )group.The levels of serum alkaline phosphatase (ALP) and urinary pyridinoline/creatinine (Pyr/Cr) were measured.Results In contrast with the Sham group,OVX group and LNG group showed significant decrease in BMD in vivo of the whole body and lumbar vertebrae as well as in BMD in vitro of whole femoral and femoral regions of interest(except for the 3rd region of interest) and whole tibia and the 1st region of the proximal tibia metaphysis ( P<0.05~P <0.01);but the serum ALP and the urinary Pyr/Cr elevated, and there was significant difference in the latter ( P <0.05). In contrast with OVX group,LNG+NYL group and NYL group showed increase in BMD in vivo and vitro, and there was significant difference in many sites( P <0.05~ P <0.01),BMD in LNG+NYL group and NYL group was close to that in Sham group.Compared with OVX group,the serum ALP and urinary Pyr/Cr in LNG+NYL group and NYL group decreased( P <0.05~ P <0.01).Conclusions LNG implant alone can not prevent the bone loss of OVX rats;NYL implant can protect the bone mass of OVX rats.
