In Vitro Effects of IFN-γ/IL-4 on Human Chorionic Trophoblast Cells and Decidual Cells in Early Pregnancy

摘要 探讨Th1/Th2型细胞因子对人孕早期绒毛滋养层细胞和蜕膜细胞活性及其内分泌功能的影响。绒毛滋养层细胞和蜕膜细胞活性采用MTT法进行检测 ,绒毛组织分泌的人绒毛膜促性腺激素 (hCG )和蜕膜组织分泌的催乳素 (PRL )均采用放免法 (RIA )进行分析测定。结果 :(1)一定浓度范围的Th1型细胞因子IFN γ (10~ 10 0 0ng/ml)和Th2型细胞因子IL 4 (1~ 10ng/ml)对绒毛滋养层细胞活性及其hCG分泌分别有不同程度的抑制作用 (P <0 0 1)和促进作用 (P <0 0 1) ;(2 )Th1型细胞因子IFN γ低浓度时 (1~ 10ng/ml)对蜕膜细胞活性及PRL分泌有刺激作用 (P <0 0 5 ) ,高浓度时 (10 0~ 10 0 0ng/ml)则有抑制作用 (P <0 0 1) ,Th2型细胞因子IL 4对蜕膜上述功能的调节作用与对绒毛滋养层的调节作用相似。 To investigate the effects of IFN γ/IL 4 cytokines on human chorionic trophoblast cells and decidual cells,and the endocrine function,the viabilities of these two kinds of cells were detected by means of MTT and radioimmunoassay (RIA) was applied to determine the secretions of human chorionic gonadotropin (hCG) from chorionic tissues and prolactin (PRL) from decidual tissues Results showed that dose range of IFN γ from 10~1 000 ng/ml and IL 4 from 1~10 ng/ml could inhibit or enhance the activities of chorionic trophoblast cells and decidual cells respectively The lower doses of IFN γ(1~10 ng/ml) stimulated the viability of decidual cells and also stimulated the secretion of PRL;but higher dose (100~1 000 ng/ml) of IFN γ showed an inhibitory effect The action of IL 4 on the above mentioned regulatory effect was similar to those to the chorionic trophoblasts It concludes that the Th1/Th2 type cytokines might express an important immunoregulatory effect in early pregnancy by means of the effects on the activities of chorionic trophoblast cells and decidual cells as well as their endocrine function
