摘要 作者探索了用聚丙烯酰胺凝胶电泳和聚丙烯酰胺等电聚焦电泳分析苜蓿酯酶同功酶和过氧化物酶同功酶的方法,确立了苜蓿种子幼芽酯酶同功酶和过氧化物酶同功酶的基本谱型。结果表明,聚丙烯酰胺过氧化物酶同功酶分析法适于苜蓿种子幼芽材料的分析,尤以等电聚焦电泳法最为灵敏,在苜蓿品种鉴定上有一定潜力。结果还表明甘肃各地方品种间及与陕北苜蓿间的亲缘关系十分密切,与新疆大叶苜蓿关系较远,与加拿大润布勒苜蓿亲缘关系最远。苜蓿的抗寒、抗旱性,等生态学特性与过氧化物酶同功酶和酯酶同功酶的酶带活性正相关,而与酶带的数量无关。 The methods of analysing esterase and peroxidase isoenzymes of Alfalfa (Medicago sativa L.) were developed using polyacrylamide gel electrophoresis (PAGE) and isoelectrofocusing (PI). The basic patterns of esterase and peroxidase isoenzymes of germinating alfalfa seeds were established. The results showed that the PI was the most sensitive method which had great potentialities in identification of alfalfa. lt could also be seen that the relationship among local alfalfas of Gansu was very close. Gansu alfalfa was close to Shanbei alfalfa, but far from Xinjiang big leaf Alfalfa and very different from Canadian Rambler alfalfa. The alfalfa's ecological characterizations such as resistance to coldness and drought were closly related to the activities of isoenzymes of alfalfa but no direct relation with the isoenzyme band numbers. The results presented here are useful to the further grassland ecological study using molecular biology.
