摘要
目的 建立沸水浴法去血浆蛋白质后用RP HPLC直接测定阿魏酸 (FA)的方法 ,求算出生化汤来源FA的药动学参数。方法 采用RP HPLC以甲醇 水 (含 1 %冰醋酸 ) (45∶55)为流动相 ,C1 8ODS柱 (4.6mm× 2 50mm ,1 0 μm) ,紫外检测波长为32 3nm。结果 FA在 4 .5~ 96 .0ng·mL- 1 范围内呈良好的线性关系 (r=0 .9975) ,方法平均回收率达 98.5 % ,RSD =1 .67%(n =3)。结论 与传统萃取法相较 ,此法更具灵敏、快速、精确、无毒、重现性好等特点 。
OBJECTIVE: To establish a RP-HPLC method for the determination of ferulic acid in plasma. METHODS: The blood samples were obtained after oral administration of Shenghua Tang. Boiling-water bath was used to deproteinate in plasma. C18ODS column (250 mm × 4.6 mm, 10 μm) was used with methanol-water (containing 1.0% acetic acid) (45: 55) as the mobile phase and UV detection at 323 nm. RESULTS: The assay was linear within the range of 4.5-96 ng·mL-1, r = 0. 997 5 (n = 7). The mean recovery was 98.5% with RSD = 1.67% (n = 3). CONCLUSION: Compared with the traditional extraction, the method appeared to be simple, rapid and sensitive, and can obtain the pharmacokinetic parameters of ferulic acid in rabbit after oral administration of Shenghua tang.
