摘要
目的 :探讨大鼠骨髓间充质干细胞 ( rat mesenchymal stem cells,r MSCs)体外诱导分化成神经元的能力。方法 :用 0 .2 5、0 .5、1 .0 mmol· L-1IBMX诱导传代的 r MSCs,待细胞分化后进行形态学观察 ,并用免疫细胞化学方法进行细胞鉴定。结果 :0 .5 mmol· L-1IBMX诱导细胞效果最好 ,2 d即可见有神经元样细胞出现 ,6d神经元样细胞占细胞总数的 ( 2 3.2± 2 .3) %,NSE染色阳性。未分化细胞表达 nestin,诱导 3d,阳性细胞增多 ,6d减少。结论 :体外 r MSC能扩增、传代 。
Objective: To observe whether rat bone marrow mesenchymal stem cells (rMSCs) could differentiate into neurons in vitro. Methods: Passaged rMSCs were induced by incubation with 0.25,0.5 and 1.0 mmol·L -1 IBMX,respectively. The differentiated neuron like cells were identified with inverted microscopy and immunocytochemistry. Results: The optic induce concentration of IBMX was 0.5 mmol·L -1 . Typical neuron like cells were observed as early as two days later. After 6 days, neuron like cells with typical neural cell morphology and NSE positive accounted for about (23.2±2.3)% of the total population. Untreated rMSCs expressed nestin, the nestin positive cells increased on 3 rd after being induced by IBMX, and nestin positve cells decreased on 6 th after being induced by IBMX. Conclusion:In vitro rMSCs are expanded, passaged and induced to differentiated into neuron like cells.
