多西紫杉醇对人血管肉瘤细胞株增殖的抑制作用

Inhibitory Effect of Docetaxel on the Proliferation of Human Hemangiosarcoma Cell Line

目的 :观察多西紫杉醇 ( docetaxel,DTX)对人血管肉瘤细胞株 ( ISOHAS)的增殖抑制作用。方法 :采用人血管肉瘤细胞株 ISOHAS进行培养 ,在培养液中分别加入不同浓度的 DTX,作用1、3、6和 9h后 ,观察 ISOHAS细胞形态的变化 ,并通过 Alamar Blue法测定 ISOHAS对抗肿瘤药的感受性。结果 :1 0 0μg· L-1浓度 DTX和 5 0 0μg· L-1浓度 DTX,作用 3h、6h的抑制率差异有显著性 ( P<0 .0 1 ) ;DTX作用 1、3、6和 9h时 ,5 0 μg· L-1浓度 DTX的抑制率与 1 0 0 μg· L-1DTX和 5 0 0 μg·L-1DTX的抑制率差异均有显著性 ( P<0 .0 1 )。结论 :DTX对 ISOHAS细胞具有较强的增殖抑制作用 。

Objective: To study the inhibitory effect of docetaxel(DTX) on the proliferation of human hemangiosarcoma cell line(ISOHAS). Methods: ISOHAS cells were cultured ,then various doses of DTX were added to the medium. After 1, 3,6 and 9 h,respectively, the conform ISOHAS cells change was measured that ISOHAS cells susceplibity to chemodrugs using Alamar Blue assay. Results :the inhibitory effect of DTX on the proliferation of ISOHAS cells ,at the dose of 100 ng·ml -1 and 500 ng·ml -1 , respectively, showed significant difference ( P <0.01) at 3 h and 6 h,respectively. When DTX was continuing for 1,3,6 and 9 h individually, the inhibitory rate of DTX among the dose of 50,100 and 500 ng·ml -1 was significant changed ( P <0.01) .Conclusion: DTX has high inhibitory effect on the proliferation of ISOHAS cells. It will be effective in the treatment of angiosarcoma.
