摘要
探讨细胞周期素E(cyclinE)和原癌基因c fos与幽门螺杆菌 (Hp)长期感染蒙古沙土鼠致胃癌前病变发生的关系。采用半定量反转录聚合酶链反应 (QRT PCR)及免疫组织化学技术检测Hp长期感染蒙古沙土鼠致胃癌前病变模型的胃窦粘膜中cyclinE和c fos的表达 ,并结合组织标本的病理学特点进行分析。Hp感染蒙古沙土鼠 2 5周时cyclinEmRNA水平的表达是正常对照的 2 2倍 ,4 5周时是 3 1倍 (P <0 0 1) ;2 5周时c fosmRNA水平的表达是正常对照的 2 5倍 ,4 5周时是 2 1倍(P <0 0 1)。免疫组织化学显示cyclinE及c fos在Hp感染蒙古沙土鼠 4 5周的表达显著高于正常对照组 (P <0 0 1)。结果表明在Hp长期感染蒙古沙土鼠致胃癌发生的早期有cy clinE和c
The purpose of this study was to explore the role of cyclin E and c fos in Helicobacter pylori ( H. pylori ) induced gastric precancerosis. The expression of cyclin E and c fos in mucosa of gastric antrum was determined by RT PCR and immunohistochemistry staining in Mongolian gerbil with H. pylori induced gastric precancerosis. Cyclin E mRNA levels were increased 2 2 fold 25 weeks ( P <0 01) and 3 1 fold 45 weeks ( P <0 01) after H. pylori infection compared with normal controls. C fos mRNA levels were increased 2 5 fold 25 weeks ( P <0 01) and 2 1 fold 45 weeks ( P <0 01) after H. pylori infection. Immunohistochemical staining revealed exclusive nuclear staining of both cyclin E and c fos. Furthermore, there was a sequential increase in cyclin E and c fos positive cells from normal epithelium to precancerosis. These data demonstrated that overexpression of cyclin E and c fos occurred relatively early in gastric oncogenesis.
