Abstract The Raman scattering characteristic difference between N9C4 and N9C8 of a glycosidie bond flanks in purine bases have been presented, with Laser Raman spectroscopic studies of calf thvmus DNA in fibers and solutions state respectively. This difference has been attributed to of between N3 and N7 Qf adjacent to the two covalence bonds, which possess unique gualitise respec-tively for the partial atomic charges on nucleicacid bases, and the water bridges construction in the primary hydration shell, and intramolecular hydrogen bond of the nucleotides. It can be considered that a N9C4 Raman hypochromism in DNA aqueous solution is because of distinct nature of N3 atom.
