摘要
目的 :探讨SD大鼠颌突外胚间充质细胞向成牙本质细胞的分化机制。方法 :在建立SD大鼠颌突外胚间充质细胞体外培养模型的基础上 ,用胰岛素样生长因子 1(IGF - 1)刺激外胚间充质细胞 ,采用倒置显微镜和免疫组化等方法从形态学和功能方面检测细胞的变化。结果 :在不含LIF的培养液中加入 10 0ng/mLIGF - 1培养的外胚间充质细胞 ,10d后在倒置显微镜下可见一些细胞出现单一细胞浆突起 ,似成牙本质细胞。消化后爬片经免疫组化抗DSP染色 ,部分细胞胞浆呈强阳性着色。结论 :IGF - 1可部分地诱导外胚间充质细胞向功能性成牙本质细胞分化 。
AIM:To investigate the mechanism of differentiation of rat ectomesenchymal cells to odontoblasts.METHODS:Based on the foundation of cultured model of rat ectomesenchymal cells, they were induced by 100 ng/mL insulin-like growth factor 1,and then changes of cells were observed morphologically and immunohistochemically. RESULTS:10 days after induction of IGF-1,some cells appeared long cytoplasmic processes protruding from the cell body, also strong positive expression of DSP was observed in cytoplasm by immunohistochemistry. CONCLUSION: Ectomesenchymal cells can differentiate to functional odontoblast-like cells in part, and may be one of the important cytokines during tooth development.
