幼年爪蟾突触前ACh受体对视顶盖神经元突触后膜GABA受体的调制

摘要 应用盲法膜片钳全细胞记录技术,以微抑制性突触后电流(miniature inhibitory postsynapticcurrents,mIPSCs)为指标,对敏感期内爪蟾突触前尼古丁乙酰胆碱受体(nAChR)调制视顶盖神经元突触后膜的GABAa受体的活动进行研究.结果表明,与成年神经元相比,从未成年动物神经元记录到的mIPSCs的振幅较小,下降时间较长.mIPSCs是由GABAa受体介导的.AChR激动剂(carbachol,nicotine,DMPP和cytisine等)均可使mlPSCs的频率增加,且carbachol的怍用需要Ca2+介导;而乙酰胆碱水解产物choline却无此作用;AChR竞争性拮抗剂DH-β-E能够拮抗carbachol诱发的mIPSCs频率增加,α3β4亚型AChR拮抗剂mecamyllamine也可以拮抗这一怍用,而在低浓度下对α7亚型AChR特异拮抗的MLA却没有这种拮抗作用.因此,乙酰胆碱以Ca2+依赖的方式参与了mIPSCs的突触前调制,其中可能主要涉及AChR的α3β4亚单位,但可能不涉及α7亚单位.同时还观察到,无Ca2+灌流可以导致巨形PSCs的出现,钳位电压对mIPSCs频率亦有调制作用.
