摘要
紫杉二烯合成酶是第一个被详细研究和目前所发现的最重要的紫杉醇合成代谢酶,它催化紫杉醇生物合成途径中的第一个定向的、慢速的步骤,然而该步骤并非限速步骤。文章综述了紫杉二烯合成酶的酶学特性、催化机理、cDNA克隆和表达,以及对紫杉醇合成代谢通量的影响等方面的研究进展,指出紫杉醇生物合成途径中的限速步骤位于距环化步骤较远的下游处。
Taxadiene synthase is the first known, and the most important, enzyme involved in taxol anabolism, which catalyzes the first committed, and a slow but not rate-limiting, step of taxol biosynthesis. In this paper, characterization, catalytic mechanism and cloning of taxadiene synthase and its role in the control of pathway flux were reviewed. Lately, potential rate-limiting steps lay farther down the pathway than the cyclization step were suggested.
