摘要
目的 :研究牙本质基质蛋白 - 1(Dmp - 1)及Cbfal在骨与软骨组织内的表达特点 ,探讨Dmp - 1在骨形成中的作用及机理。方法 :原位杂交检测小鼠胚胎骨发育过程中 (12 .5dpc至出生后 8周 )Dmp - 1及Cbfal在骨组织中的时空表达谱。PCR法研究Dmp - 1的表达与体外”骨小结”形成及矿化的联系。 结果 :Dmp - 1首先出现在肥大软骨细胞中 ,接着是成骨细胞 ,最后在骨细胞中强烈表达。发育过程中Cbfal和Dmp - 1在软骨和骨中重叠表达 ,且Cbfal在Dmp - 1之先。Dmp - 1的表达与鼠婴颅盖骨细胞内”骨小结”形成及体外矿化关系密切。 结论 :本研究表明Dmp - 1是成骨细胞分化和骨组织形成中的特殊标记基因 ;Dmp -
Objective:To study the characteristics of Dmp-1 and Cbfal expression in bone and cartilage, and to investigate the role of Dmp-1 in osteogenesis.Methods:In situ hybridization studies were performed to characterize the spatial and temporal expression patterns of Dmp-1 during development in mouse embryos from 12.5 dpc to 8-week postnatal.PCR was used to examine the relationship between Dmp-1 expression and bone nodule formation and mineralization in vitro.Results:Dmp-1 first appeared in hypertrophic cartilage cells, followed by osteoblasts, and later was strongly expressed in osteocytes.The expression profiles of Cbfal and Dmp-1 overlapped in both cartilage and bone during development,with Cbfal preceding Dmp-1.Dmp-1 expression was closely associated with bone nodule formation and mineralization in fetal rat calvarial cell cultures.Conclusions:These studies hilight Dmp-1 as a unique marker gene for osteoblastic differentiation and bone morphogenesis. Both Dmp-1 and Cbfal may play an important role in bone development.
