Effect of valsartan on scavenger receptor mRNA expression in human glomerular mesangial cells

摘要 目的探讨缬沙坦对人肾小球系膜细胞(HMC)表达清道夫受体(SR)水平的影响。方法应用流式细胞仪和逆转录.聚合酶链反应(RT-PCR)检测体外培养HMC在佛波酯(PMA)刺激后缬沙坦对HMC SR的mRNA的表达。结果体外培养的HMC在PMA刺激下能大量表达SR mRNA,呈剂量依赖性,其基因序列测定结果与巨噬细胞所表达的SR完全相同。缬沙坦能抑制PMA诱导的HMC SR表达,且呈剂量及时间依赖性。结论HMC具有表达SR的潜力,缬沙坦能明显抑制HMCSR在PMA诱导下的表达,这可能是其发挥治疗作用的机制之一。 Objective To investigate the effect of valsartan on expression of scavenger receptor (SR) in cultured human mesangial cells (HMC) . Methods Flow cytometry analysis and RT-PCR were used to detect the expression of SR mRNA in HMC cultured in media after the induction of phorbol 12-myristate 13-acetate(PMA) with or without valsartan. SR was sequenced and compared with the data of human macrophage gene bank. Results SR mRNA in HMC was up-regulated in dose-dependant manner after PMA treatment. The RT-PCR products were the same as the sequence of SR mRNA in macrophages. Valsartan could down-regulate SR mRNA expression in dose- and time-dependant manner. Conclusions SR can be expressed in HMC and will up-regulate in case of PMA treatment. Valsartan can down-regulate the SR mRNA expression markedly.
