摘要
目的 建立类似于人类干燥综合征的实验动物模型 ,模拟其唾液腺的病理改变和口腔临床表现 ,并试图就腺体的淋巴细胞浸润程度、饮水量及口干症状与时间的关系进行研究。方法 运用BALb C近交系小鼠颌下腺组织匀浆与弗氏完全佐剂(CFA)混匀多点注射小鼠皮下 ,再注射百日咳疫苗加强免疫 ,产生类似于人类干燥综合征 (SS)的唾液腺改变及临床表现。结果 给BALb C小鼠注射同种颌下腺抗原后 ,其颌下腺淋巴细胞浸润的程度不一 ,2、4、6周淋巴细胞浸润有逐步加重的趋势 ,但前两组饮水量及唾液流率无显著性差异 ,6周组的损害最重 ,饮水量增加而唾液流率却显著减少。结论 初步建立了类似于人类SS表现的动物模型。并证实腺体的淋巴细胞浸润程度、饮水量及口干症状与时间密切相关。
Objective To develope a mouse model for non bacterial sialoadenitis similar to Sjgren's syndrome and try to find out the relationship between lymphocytic infiltration ,water drinking test, symptoms of dry mouth and time. The model of organ specific to the submandibular gland of BALb/C mouse has developed for this study. Methods 60 BALb/C inbred line mice were divided into three groups(2?4?6week),in each group 10 were injected by homologous antigen of submandibular gland tissues mixed with completely freund's adjuvant(CFA), pertussis vaccaine injected to enhence immuological reaction. another 10 mice were injected only by the same volume saline as control experiment, submandbular glands were analyzed histologically at different time points. Results 2?4?6weeks later mice in each groups were killed and submandbular lopes were excised and snaped frozen for frozen sections, Lesions of submandbular glands in 6 week group are more severer than that in the other two groups, but the first two groups have no significant differences.Conclusion A mouse model of non bacterial sialoadenitis has been set up and the lesions may be similar to the autoimmune progress in human Sjgren's syyndrome. This study indicates the degree of lymphocytic infiltration,and clinical symptoms could be much severe with time passed .
