摘要 用高效液相色谱法 (HPLC)在半制备硅胶柱上将大豆磷脂分离成各类磷脂 ,柱后收集磷脂酰乙醇胺(PE) ,磷脂酰胆碱 (PC) ,磷脂酰肌醇 (PI) ,以甲醇 氯仿 乙腈 水为流动相 ,用梯度洗脱程序在ProdigyODS(3 )柱上分离分子种 ,PE、PI和PC各被分离成 9个、10个和 11个分子种。组分定性由HPLC峰的脂肪酸组成确定。此方法溶剂易得 ,简单快速 (仅需 3 0min) ,可用于分离各种大豆磷脂分子种。 Soybean phospholipids were separated into phospholipids on a semi-preparative s ilica gel column using HPLC. Phosphatidylcholine(PC), phosphatidylethanolami ne(PE) and phosphatidylinositol(PI) fractions were collected. Their molecular sp ecies compositions were analysed on a revers phase Prodigy ODS(3) column and a g radient elution program performed with chloroform-methanol-acetonitrile-water as a flow phase. Phosphatidylcholine, phosphatidylethanolamine and phosphatidyl inositol were separated into 11, 9 and 10 molecular species respectively. The molecular species identities were assign ed from fatty acid compositions determined by gas chromatography(GC) . The method has advantages of simplicity and availability of solvents .
