人参皂甙Re抗大鼠急性缺血-再灌流心肌细胞凋亡及相关基因蛋白表达 被引量:20

Effects of ginsenoside Re-on anti-cardiomyocyte apoptosis and the expression of related gene proteins in rats after acute ischemia and reperfusion

摘要 目的 观察人参皂甙Re抗急性缺血再灌流心肌细胞凋亡及Bcl 2、Bax、Bad和Fas基因蛋白表达 ,探讨人参皂甙Re抑制心肌细胞凋亡的可能机制。方法 结扎Wistar大鼠左冠状动脉前降支 ,建立大鼠缺血 再灌流动物模型 ;采用透射电镜、缺口末端标记法检测心肌凋亡细胞 ,利用光学显微镜进行细胞计数 ;免疫组织化学检测Bcl 2、Bax、Bad和Fas基因蛋白的表达 ,并利用图像分析系统测量平均光密度值进行定量分析。结果 ①假手术组未发现心肌凋亡细胞。缺血 再灌流组心肌凋亡细胞数为 (13 4 4 5± 4 5 61)个 /视野 ,Re治疗组 (90 66± 19 2 2 )个 /视野 ,两组间差异有显著性意义 (P <0 0 1)。②免疫组织化学检测发现缺血 再灌流组及Re治疗组Bcl 2、Bax、Bad和Fas基因蛋白的表达较假手术组明显增加 (P <0 0 5 ) ,Re治疗组Bcl 2的表达与缺血 再灌流组比较差异无显著性意义 (P >0 0 5 ) ,而Bax、Bad、Fas的表达明显下降 (P <0 0 5 ) ,人参皂甙Re治疗组Bcl 2 /Bad、Bcl 2 /Bad、Bcl 2 /Fas比值均较假手术组与缺血 再灌流组明显增大。结论 人参皂甙Re治疗可以抑制急性缺血再灌流诱导的心肌细胞凋亡 ,其作用机制可能是抑制了促凋亡基因Bax、Bad、Fas的表达 ,从而使Bcl 2 /Bax、Bcl 2 /Bad、Bcl 2 /Fas比值增大。 Objective To observe the effects of ginsenoside Re on anti cardiomyocyte apoptosis and the expression of Bcl 2,Bax,Bad,and Fas gene proteins in rats after acute ischemia and reperfusion ,and to explore the possible mechanisms of ginsenoside Re on inhibition of cardiomyocyte apoptosis.Methods The ischemia reperfusion heart model was established by ligating the left anterior descending branch of coronary artery in Wistar rats.The apoptotic cardiomyocytes were measured with transmission electron microscopy and counted with in situ nick end labeling(TUNEL) method and light microscopy.The expression of Bcl 2,Bax,Bad,and Fas gene proteins were studied by immunohistochemical staining.Mean optical density(OD) value of the gene proteins expression were quantitatively examined by image analysis system.Results ①The apoptotic cardiomyocytes didnt exist in sham operation group.The number of the apoptotic cells was(134 45±45 61)per field in the ischemia/reperfusion group,and (90 66±19 22)per field in the ginsenoside Re treated group.The difference was significant among the later two groups( P <0 01).②The expression of Bcl 2,Bax,Bad,and Fas gene increased significantly in the ischemia/reperfusion group and ginsenoside Re treated group,compared with the sham operation group( P <0 05).There were not significant difference in the Bcl 2 gene expression between the ginsenoside Re treated group and ischemia reperfusion group( P >0 05),but the expressions of Bax,Bad,and Fas gene decreased significantly in the ginsenoside/Re treated group,compared with the ischemia/reperfusion group( P <0 05).Compared with the ischemia/reperfusion group and the sham operation group,the ratios of Bcl 2/Bax,Bcl 2/Bad,and Bcl 2/Fas rised significantly in the ginsenoside Re treated group.Conclusion Ginsenoside Re can significantly inhibit cardiomyocyte apoptosis induced by ischemia/reperfusion in rats.The mechanism is that ginsenoside Re inhibits the expressions of pro apoptotic Bax,Bad,and Fas gene and raises the ratios of Bcl 2/Bax,Bcl 2/Bad,and Bcl 2/Fas.
