尿毒净Ⅱ号对人肾系膜细胞增殖影响的实验研究

Effects of niaodujing Ⅱ on the proliferation of human glomerular mesangial cells in vitro

目的 :探讨尿毒净Ⅱ号对人肾小球系膜细胞增殖的影响。方法 :体外培养人胚胎肾小球系膜细胞 ,应用氮蓝四唑盐 (MTT)比色法、流式细胞术对DNA倍体进行分析 ,分别检测尿毒净Ⅱ号对系膜细胞增殖的影响。结果 :尿毒净Ⅱ号明显抑制血清加内皮素 1(ET 1)刺激的系膜细胞增殖 (P <0 .0 1) ,G0 G1期细胞比例增加 ,S期细胞比例逐渐下降 ,呈一定的量效和时效依赖关系。结论 :尿毒净Ⅱ号通过抑制细胞DNA合成 ,延缓了细胞周期进程 ,从而抑制了系膜细胞增殖。

AIM: To investigate the effects of niaodujing Ⅱ on cultured human glomerular mesangial cells. METHODS: The proliferation of cultured mesangial cells (MsC) in human embry, stimulated by serum and ET-1 following incubation with niaodujing Ⅱ, was measured in vitro by MTT method and flow cytomertry (FCM). RESULTS: Niaodujing Ⅱ inhibited the proliferation stimulated by serum and endothelin-1 (ET-1) in a dose-dependent and time-dependent manner (P< 0.01). In the mesangial cell cycle, the number of cells in G 0/G 1 phase increased and the number in S phase decreased. CONCLUSION: Niaodujing Ⅱ can suppress the proliferation of MsC by inhibiting DNA synthesis.
