目的 探讨PTEN基因在胰腺癌组织中的表达及其临床意义。方法 应用免疫组化Envision法检测 5 4例胰腺癌和10例正常胰腺组织中PTEN蛋白的表达情况 ,并分析与胰腺癌临床病理因素的关系。结果 PTEN蛋白在胰腺癌组织中总的表达率为 3 1.5 % (17/ 5 4) ,在Ⅱ、Ⅲ及Ⅳ期胰腺癌组织中的表达率分别为 5 0 .0 % (9/ 18)、2 7.3 % (6/ 2 2 )和 14 .3 % (2 / 14 ) ;有远处转移与无远处转移的胰腺癌组织中PTEN蛋白表达率分别为 2 0 .0 % (2 / 10 ) ,3 4.1% (15 / 44) ;有淋巴结转移与无淋巴结转移PTEN蛋白表达率分别为 18.2 % (4 / 2 2 ) ,40 .6% (13 / 3 2 )。PTEN蛋白在G1、G2 和G3 级胰腺癌组织中的阳性率分别为 60 .0 % (6/ 10 )、2 8.0 % (7/ 2 5 )和 2 1.1% (4 / 19)。 10例正常胰腺组织PTEN阳性表达率为 70 .0 % (7/ 10 )。结论 胰腺癌的分期愈晚 ,PTEN蛋白阳性表达率愈低 ;分化程度愈低 ,表达愈低。PTEN蛋白的阳性表达对胰腺癌的发生 ,发展起着重要作用 。

Objective To investigate the expression of PTEN in pancreatic tissues and it′s clinical significance.Methods A total of 54 pancreatic cancers were studied for PTEN expression via immunohistochemistry.10 normal tissues of pancreas were used as control.Results The overall expression rate of PTEN in pancreatic cancers was 31.5%(17/54),while the positive rate of PTEN expression was 50.0%(9/18),27.3%(6/22) and 14.3(2/14) in stage Ⅱ,Ⅲ,Ⅳ of pancreatic cancers respectively.The ratio of positive expression of PTEN was 20.0%(2/10) and 34.1%(15/44) in pancreative cancer tissues with distant metastasis and without distant metastasis;The ratio of positive expression of PTEN was 18.2%(4/22) and 40.6%(13/32) in pancreative cancer tissues with lymph node metastasis and without lymph node metastasis.The positive rate of PTEN in histological grade Ⅰ,Ⅱ,Ⅲ was 60.0% (6/10),28.0%(7/25) and 21.1(4/19) respectively.PTEN was found in 7 of 10 normal pancreatic tissues,and its positive rate was 70.0% (7/10).Conclusion PTEN is negatively correlated with the staging of pancreatic cancer as well as with the differentiation of pancreatic cancer,it plays an important role in pancreatic cancinogenesis.
