摘要
目的 :检测白血病细胞系红细胞生成素受体 (Epo R)及 H- Ras的表达 ,阐明 Epo R/H- Ras介导的白血病细胞系 HL- 6 0、K5 6 2增生信号传导途径。方法 :3H- Td R掺入法检测各细胞系对Epo刺激的增生反应 ;Western印迹法检测 HL- 6 0、K5 6 2细胞系中 Epo R、H- Ras表达。结果 :1HL-6 0、K5 6 2细胞系 Epo R表达的阳性率分别为 6 6 %和 6 1%。 2 HL- 6 0、K5 6 2细胞系受 Epo刺激而显著增生 ,同时伴有 H- Ras显著增高 ,P<0 .0 0 1。结论 :白血病细胞系可有 Epo R、H- Ras的表达 ;Epo可通过 Epo R/ H-
Objective:Methods:To investigate the expression of erythropoietin receptor (EpoR) and H Ras in leukemic cell lines,HL 60?K562,and to clearify the mechanism of proliferative signal transmission in the leukemic cell mediated by EpoR.Results:Biotinylated Epo and flow cytometry were used for EpoR expression, 3H TdR incorporation for cell proliferation, Western blot analysis for the expresson of H Ras.Conclusion:The cell proliferation could be stimulated by Epo in some leukemic cell lines, modulated by EpoR/H Ras pathway.Either of the two cell lines expressed high level of EpoR,with the positive rate as 66 %?61 %,respectively. The cell proliferation was significantly enhanced in response to Epo in the two cell lines, accompanying with higher lever of H Ras expression.
