气虚证模型大鼠鼻咽组织cDNA阵列C区基因表达谱特征及其对治疗的反应 被引量:11

摘要 目的探讨气虚证模型大鼠鼻咽组织C区基因谱表达特征及其对治疗的反应。方法药物加疲劳法在Wistar大鼠制作气虚模型,随机分为治疗组与不治组,以AtlasTM cDNA阵列法检测造模各组动物鼻咽组织C区基因表达谱,比较分析其表达谱差异。结果模型组动物表现蛋白激酶B(PKB)、甘丙肽(galanin)前体、钙泵(PMCA)基因表达下调。治疗一周后,表达下调的PKB和PMCA基因活性恢复正常,但galanin基因活性仍维持下调状态。结论单纯气虚证动物鼻咽组织凋亡相关蛋白基因表达活性下调,其凋亡信号传导通路发生异常。经针对性治疗后,这些异常在一定时间内可以恢复正常。此类变化趋势和病理生理意义在于尽力维持机体内环境的稳定,即阴阳平衡。 Objective To investigate the features of gene expression profiling on C area of Atlas?cDNA array and their response to treatment in the nasopharyngeal tissues of rats with Qi deficient syndrome. Methods Qi deficient syndrome was induced in rats with the methods of herbal decoction perfused gastricaly and supplemented by forced swimming in deep pool. Then, the expressive features of genes on C area were determined in nasopharyngeal tissues of tested rats with Atlas?cDNA expression array. Results The expressive activities of protein kinase B (PKB), galanin precursor(GALN) and calcium-transporting ATPase(PMCA) were down regulated among the rats of modeling group, while GALN was the only gene remaining in an abnormal status of activity down regulated among the rats of treating group. Conclusion The features of gene expression profile with rats of Qi deficient syndrome were the abnormal expression activities of apoptosis-related protein genes, mainly in a status of down regulating, and these down regulated gene activities can be restored to a normal expressing status after treatment with appropriate therapy. The implications of this pathological process lie in the maintenance of the balance of Yin and Yang within the diseased body to a most favorable degree.
