摘要
同步辐射源产生的X射线具有高通量、高准直性及波长连续可调等优点,应用于结构生物学将可以解决很多常规实验装置无法解决的问题.文章介绍了同步辐射X射线衍射装置及其在结构生物学中的应用,蛋白质晶体学中相位问题的有关进展,以及国家同步辐射实验室(NSRL)二期工程X射线衍射与散射光束线、实验站的实验装置及最新的研究进展.
Many unsolvable problems can be tackled when synchrotron radiation X\|rays are applied with their superior advantages of high flux, high collimation, good polarization and tunable wavelength. We present an overview of our synchrotron radiation X\|ray diffraction facility and its application in macromolecular crystallography.We also review the progress in solving the phase problem of protein crystals. Finally, we give a short description of the construction of the X\|ray diffraction and scattering beamline and station, as part of our laboratory′s Phase Ⅱ project.
