PGE_2对胰腺癌PC-3细胞株血管内皮生长因子(VEGF)表达的影响

目的 探讨PGE_2对胰腺癌PC-3细胞株血管内皮生长因子(VEGF)表达的影响。方法 应用逆转录-聚合酶链反应(RT-PCR)和酶联免疫吸附测定(ELISA)方法,在选择性环氧合酶(COX)-2抑制剂Celebrex的干预的基础上,观测PGE_2对胰腺癌PC-3细胞株VEGF表达的影响。结果 PGE_2可上调胰腺癌PC-3细胞株VEGF的表达,且表现出一定的剂量依赖性关系。结论PGE_2可上调胰腺癌PC-3细胞株VEGF的分泌,其可能在COX-2参与胰腺癌新生血管生成的过程中起着重要的介导作用。

Objective To investigate the effects of PGE2 on the expression of vascular endothelial growth factor (VEGF) in pancreatic carcinoma PC-3 cell lines. Methods Based on the interference of the selective COX-2 inhibitor, Celebrex, to explore the influence of PGE2 on the expression of VEGF in pancreatic carcinoma PC-3 cell lines by using reverse transcription polymeraae chain reaction ( RT-PCR) and enzyme-linked immuno-absordent assay(ELISA). Results The expression of VEGF was up-regulated by PGEj in a dose-dependent manner in pancreatic cancinoma PC-3 cell lines. Conclusion PGE2 can up-regulate the expression of VEGF in pancreatic carcinoma PC-3 cell lines, and it may pky an important intermediate role in the process of angiogenesis together with COX-2.
