摘要
目的:采用RP-HPLC法检查羟喜树碱的有关物质。方法:采用Diamonsil C_(18)(200mm×4.0mm,5μm)色谱柱,流动相:甲醇—水(40:60);检测波长分别为:254nm、266nm、373nm(喜树碱和羟喜树碱的最大吸收波长和两者等吸收系数点波长);柱温:40℃;流速:0.8ml/min。结果:254nm波长处:喜树碱在0.536μg/ml~5.36μg/ml浓度范围內呈良好的线性关系,回归方程Y=5695.7C+56.3,相关系数r=0.9995,最低检测浓度:0.268μg/ml最低检测量,5.36ng。结论:方法简便,快速,准确。可定量测定羟喜树碱中的主要杂质喜树碱。
Objective:To establish RP-HPLC method for the test of the related substances of Hydroxycamptothecin. Methods: The chromatographic conditions included column Diamonsil C18(200mm×4.0mm5um),mobile phase of methanol water(40 : 60)and detector UV 254 nm, 266 nm and 373 nm ,the column temperature was 40℃,the flow rate was 0. 8 mL/min. Results : There was a good linear relationship within the concentration range of 0. 536ug/mL^5. 36ug/mL of Camp-to thecin at 254nm. The regression equation : Y = 5695. 7C + 56. 3 ,r = 0. 9995. Conclusion: This method is simple . quick and accurate for the test of the main substances (Camptothecin)of Hydroxycamptothecin.
