加味芍甘提取物对高催乳素血症小鼠激素水平的影响

Effect of Extract of Flavored Shaogan on Hormone Level in Hyperprolaetinemic Mice

目的观察加味芍甘提取物对高催乳素血症(HPRL)模型小鼠激素水平的影响。方法将小鼠随机分成10组:正常对照组、模型组、模型对照组、溴隐亭组、加味芍甘醇提取物低、中、高剂量组、加味芍甘水提取物低、中、高剂量组。皮下注射甲氧氯普胺注射液制作小鼠HPRL模型,给药30 d后,采用酶联免疫分析法测定小鼠血清催乳素(PRL)、雌二醇(E2)及孕酮(P)水平,并称量小鼠子宫质量,计算子宫指数。结果模型组和模型对照组小鼠血清PRL水平升高,血清E2、P水平降低,与正常对照组比较有显著性差异(P<0.01);加味芍甘醇提物与水提物的高、中、低剂量及溴隐亭均能不同程度的降低血清PRL水平,升高血清E2、P水平及子宫指数,与模型对照组比较有显著性差异(P<0.05或P<0.01)。随着醇提物剂量的增加,小鼠PRL含量逐渐下降,E2含量及子宫指数逐渐升高;水提物中剂量降低小鼠PRL水平、升高E2、P水平作用强于高、低剂量组。结论加味芍甘汤的醇提取物与水提物能降低HPRL小鼠血清PRL,促进子宫发育,调节性激素水平。加味芍甘水提物具有双向调节作用,其中剂量功效与溴隐亭相当。

Objective To study the effect of the extract of flavored shaogan in hyperprolaetinemic( HPRL) mice.Methods Metoclopramide was injected into mice to establish the animal HPRL model. Mice were randomly divided into10 groups: normal group; model group,model control group; bromocriptine group; low,medium and high doses of alcohol extract groups; low,medium and high doses of water extract groups. The intervention period lasted for 30 days. Serum PRL,E2,P levlels were determined with ELISA method,respectively,in mice. The uterus of mice was weighed and the index was calculated. Rusults Serum PRL level was increased,while E2,P levels decreased in model group and model control group( P < 0. 01 compared with normal group). Serum PRL level was decreased,E2 and P levels were increased to some extent after the administration of the extract of flavored shaogan,while the uterus index was elevated( P < 0. 05 or P < 0. 01 compared with model control group). The dosage of alcohol extract was positively related to the effects on hormone levels. With the medium dose of water extract,the PRL level was decreased,while E2 and P levels were increased more significantly than that in the high and low dose groups. Conclusion The alcohol extract and water extract of flavored shaogan soup can decrease serum PRL,promote the development of uterus,and regulate the hormone level in HPRL mice. The water extract has a dual-direction regulation effect and the medium dose has a similar effect to bromocriptine.
