摘要
采用薄层扫描法(TLC),以正己烷 氯仿 无水乙醚 冰醋酸(体积分数比为4∶3∶2∶0 1)为展开剂,碘蒸气为显色剂可以同时测定发酵液中阿魏酸、香草酸和香草醛的含量.用2,4 二硝基苯肼显色可以专一性地测定香草醛.用碘蒸气显色时阿魏酸、香草酸和香草醛的线性范围均为1~10μg,标准曲线分别为y=353 3x-183 2,R2=0 9954;y=308 0x-42 0,R2=0 9979;y=655 11x-49 0,R2=0 9954.平均回收率分别为101 3%,101 8%,103 0%,RSD分别为2 39%,2 39%,2 24%.用2,4 二硝基苯肼显色时香草醛的灵敏度更高,线性范围为1~5μg,标准曲线分别为y=6843 6x-4776,R2=0 9922.两种显色方法测得的香草醛结果一致.
The contents of ferulic acid,vanillic acid and vanillin in fermentation liquid can be determined simultaneously by TLC,with nhexanechloroformdehydrated etherglacial acetic acid (4∶3∶2∶01) as devekoper and iodine vapor as colordeveloping agent.Vanillin can be determined with 2,4nitrophenyhydrazine as colordeveloping agent specially.The linear range of feruli acid,vanillic acid and vanillin are all between 1~10 μg,their standard curve are y=3533 x-1832,R2=0995 4;y=3080 x-420,R2=0997 9;y=65511 x-490,R2=0995 4 respectively.The average recovery are 1013%,1018% and 1030% respectively,RSD are 239% and 224%.The method of using 2,4nitrophenyhydrazine is more sensitive.The linear range is 1~5 μg,and the standard curve is y=6 8436 x-477 6,R2=0992 2.The determination result of vanillin is same by these two methods.
