γ-射线诱发的人AHH—1淋巴细胞凋亡及其机制研究

摘要 研究γ-射线诱发的人AHH-1淋巴细胞凋亡规律和发生机制,用MGG、TUNEL染色,MTT、免疫组化和电镜观察,经0、3、6、9、12、15、20Gy γ-射线照射后,AHH-1淋巴细胞凋亡、活存率以及Bax、Bel-2、Bel-xl等凋亡相关蛋白表达的变化。结果(1)3—12Gy照射范围内,凋亡是AHH-1细胞死亡的主要方式,且6-12Gy范围内呈现较好的量效关系;(2)15-20Gy照射后,凋亡率下降,推测此时除凋亡外,坏死也是AHH-1细胞死亡的主要途径;(3)不同剂量照射后,AHH-1细胞凋亡率在24h达到高峰;3-12Gy照射范围内,细胞活存率随照射剂量的增加而下降,15-20Gy照射后,下降趋势趋于平缓。结论认为一定剂量γ-射线照射可诱发人AHH-1淋巴细胞出现大量凋亡,且在一定范围内呈现较好的量效关系。 To investigate the apoptosis and its mechanism of human lym-phoblast cell line (AHH-1) induced by γ-ray, using TUNEL assay, May-Grunwald-Gimsa staining, MTT, immunohistochemical technique and TEM,the apoptosis, survival rates and the expression of Bax, Bcl-2 and Bcl-xl pro-teins of AHH-1 irradiated by 0, 3, 6, 9, 12, 15 and 20 Gy γ-rays are stud-ied. It is found that (1) Apoptosis was the major pathway of lymphocyte deathwithin 3-12 Gy, and showed a good dose-effect relationship within 6--12Gy. (2) The rate of apoptosis dropped when the doses of 15-20 Gy were de-livered. It was suggested that necros might the another pathway of AHH--1lymphocyte death besides the apoptosis. (3)At 24 h after different doses irra-diation, the apoptosis of AHH-1 lymphocyte reached the maximal. The cellsurvival decreased evidently within 3--12 Gy and tended to be slow after 15-20 Gy radiation. It is concluded that γ-ray can induces mass apoptosis ofAHH-1 lymphocyte and shows a good relationship within a definite doserange.
