摘要
为了获得高纯度茶多酚及表没食子儿茶素没食子酸酯(EGCG),开发了一种可连续化操作的中压交联聚乙烯基吡咯烷酮(PVPP)柱层析纯化茶多酚及EGCG的方法。在静态吸附解吸实验的基础上,确定了连续中压柱层析分离的最佳工艺条件,即1 500mm×200mm的自制不锈钢中压层析柱,洗脱液为丙酮,洗脱流速为30mL/min。提取液经上述分离纯化操作后,产品中w(茶多酚)>98%,w(咖啡因)<1%,w(EGCG)由原来的17.16%提高至60.12%,收率为85.25%。以PVPP为填料,丙酮为洗脱液的操作,缩短了生产周期,降低了生产成本,具有良好的工业化应用前景。
The purification of tea polyphenols(TP)and epigallocatechin gallate(EGCG)by successive medium-pressure polyvinylpyrrolidone(PVPP)column chromatographic extraction(CCE)was studied.On the base of static absorption and desorption experiments,the optimum conditions of CCE were determined,that is,1 500mm×200mm stainless steel column was used as chromatographic column,PVPP as filling material,acetone as eluting solvent,flowing rate of 30mL/min.The content of EGCG was increased to 60.12%from17.16%and the yield of it was 85.25%after the purification.Moreover,the content of TP was more than 98%and the content of caffeine was less than 1%.The technical process simplified the operations,prevented the environmental pollution and lowered the costs.
