姜黄素调节Bcl-2、Bax蛋白表达诱导子宫内膜癌(HEC-1-B)细胞凋亡

摘要 目的:研究姜黄素诱导人子宫内膜癌(heC-1-B)细胞凋亡的分子机制。方法:将体外培养的heC-1-B细胞随机分5组,对照组、姜黄素i、姜黄素ii、姜黄素iii、姜黄素iV。分别给与不同浓度的姜黄素(0,10,20,40,80)μmol/l处理heC-1-B继续培养48h、72h。mTT法检测姜黄素对细胞增殖的影响,WeSTern BloT法检测BCl-2、Bax蛋白表达。结果:mTT结果显示姜黄素对(heC-1-B)细胞具有抑制作用,抑制率随姜黄素浓度和作用时间的增加而上升(p<0.05);WeSTern BloT结果显示BCl-2蛋白表达下降,Bax蛋白表达升高,Bax/BCl-2比值增大,差异有统计学意义(p<0.05)。结论:姜黄素对(heC-1-B)细胞有抑制作用并能诱导其凋亡,其机制与下调BCl-2蛋白表达、上调Bax蛋白表达有关。
