黄芪多糖不同干预条件下对高糖刺激小鼠肾小球足细胞Nephrin表达的影响

摘要 目的:体外模拟高糖病理微环境,探讨黄芪多糖对小鼠肾小球足细胞Nephrin表达的影响。方法:试验一:体外培养小鼠肾小球足细胞,分别为D-葡萄糖30mmol/L+黄芪多糖干预组(0.0g/L、0.1g/L、0.2g/L、0.4g/L和0.8g/L);干预时间为72h。试验二:分别为D-葡萄糖30mmol/L+黄芪多糖干预(浓度为0.X g/L),干预时间分别为0h、3h、6h、12h、24h、48h和72h。分别采用流式细胞计数检测各组足细胞Nephrin表达。结果:随着黄芪多糖干预浓度的递增,足细胞表面Nephrin表达逐渐上调,呈浓度依赖性(P<0.05);随着黄芪多糖干预时间的延长,足细胞表面Nephrin表达逐渐上调,呈时间依赖性。结论:在高糖刺激下,黄芪多糖干预可上调肾小球足细胞Nephrin表达,是黄芪治疗糖尿病肾病蛋白尿的可能作用靶点。
