日本黑松组织培养初步研究

Preliminary Study on Tissue Culture of Pinus thunbergii

以日本黑松针叶束为外植体试材进行组织培养,探讨消毒试剂和时间、基本培养基、外源激素的种类和使用浓度、蔗糖浓度等对日本黑松组织培养的影响,结果表明:选用70%的酒精30 s和0. 1%HgCl_2 10 min对外植体消毒效果最佳、SH培养基附加6-BA0. 5 mg·L^(-1)+NAA 0. 05 mg·L^(-1)+30 g·L^(-1)蔗糖+6 g·L^(-1)琼脂时诱导愈伤组织效果最好,愈伤组织诱导率为58. 00%。

Tissue culture of Japanese black pine needle bundles was chosen as explants was carried out to investigate the effects of disinfection reagents and time,basic medium,types and concentrations of exogenous hormones as well as concentration of sucrose on tissue culture of Pinus thunbergii. The result showed that 70 % alcohol 30 s and 0. 1 % HgCl2 10 min had the best disinfection effect on explants. The effect of callus induction by using SH medium supplemented with 6-BA0. 5 mg·L-1+ NAA 0. 05 mg·L-1+ sucrose 30 g·L-1+ agar 6 g·L-1 was the best,and the callus induction rate was 58. 00 %.
