摘要
目的 :探讨肌酐的毛细管区带电泳多次进样技术。方法 :以肌酐、吡啶为例 ,采用 3 6cm× 5 0 μm(i .d)涂层柱 ,0 .1mmol/LpH2 .5的磷酸缓冲液 ,在线多波长检测。多次进样技术是采用改变应用电压、电泳时间两种模式。 结果 :用电压控制模式 ,连续进样五、六个样品总分析时间分别为 10min、11min ,比单次进样分别节省 15min、19min左右 ;采用内标法定量及 5次进样技术 ,肌酐与吡啶的峰高比值的变异系数小于 6.0 % ,且在肌酐浓度为 5~ 80mg/L范围内 ,肌酐浓度与肌酐同吡啶的峰高比值的线性关系非常好 (r =0 .9996)。结论 :多次进样技术采用电压控制模式简便有效。最大进样次数为单次进样分析中诸相邻峰分离度中最小值 (取整数 )。
Objective:To study multiple injection technique of capillary zone electrophoresis (CZE) for creatinine detection.Methods:The effect of multiple injection technique of CZE for detecting creatinine and pyridine was observed with voltage or time control model.Results:The voltage control model was a more conveient,effective and simple technique in comparison with the time control model.Compared with that of single injection,the total time of 5 or 6 successive injections in the voltage control model was cut down by 15 or 19 minutes ,respectively.Using 5 successive injections and internal standard,the variant coefficient of the peak ratio between creatinine and pyridine was less than 6.0%,and the linear relation between creatinine concentration and the peak ratio was good ( r =0.9996)at strengths of 5~80 mg/L of creatinine.Conclusion:The voltage control model could be very suitable for multiple injection technique of CZE
