肝硬变门静脉高压症大鼠肝组织内皮素-1基因的mRNA表达研究

Expression of endothelin-1 gene at the level of mRNA in hepatic tissues of cirrhotic rats with portal hy- pertension

目的研究肝硬变大鼠肝组织内皮素-1(ET-1)的基因 mRNA 表达的变化。方法半定量逆转录多聚酶链反应(SqRT-PCR)。结果经 SqRT-PCR 测定大鼠肝组织 ET-Ⅰ基因的 mRNA(integral optical den-sity,IOD)值:各期肝硬变大鼠模型制作2周、4周、6周、8周分别为68330±1096、73836±1125、85712±1283、89215±1366,均明显高于正常对照鼠37745±1066(P<0,01),但肝硬变大鼠各组之间的差异无统计学意义(P>0.05)。结论肝硬变门静脉高压症时大鼠肝组织内 ET-1基因的 mRNA 表达明显增加。

Objective To make semiquantitative analysis of mRNA expression of endothelin-l (ET-l) gene in the hepatic tissues of rats with portal hypertension.Methods Semiquantitative reverse transcription and poly- merase chain reaction (SqRT-PCR) was used.Results Expression of ET-l gene at the level of mRNA in hep- atic tissues by SqRT-PCR(integral optical density) in various period after induction of the cirrhosis,i.e.week 2,4,6,and 8 was significantly higher in cirrhotic groups;68330±1096,73836±1125,85712±1283 and 89215±1366 than in normal control groups (37745±1066)(P<0.01).Differences between cirrhotic groups were not significant(P>0.05).Conclusion Expression of mRN A of ET-l gene in hepatic tissues is significantly higher in cirrhotic rats than in normal rats.
