摘要
目的 :探讨大鼠心肌缺血损伤与凋亡相关基因bcl 2和bax表达的关系及牛磺酸的影响。方法 :结扎大鼠冠状动脉左前降支建立心肌缺血模型。检测心肌线粒体中超氧化物歧化酶 (SOD)、Ca2 + ATPase活性及MDA(丙二醛 )含量 ,用免疫组化法检测Bcl 2和Bax蛋白在心肌中的表达量。结果 ;结扎冠脉左前降支可致心肌线粒体MDA含量升高 ,SOD和Ca2 + ATPase活性下降 ;缺血心肌Bax蛋白表达呈显著升高 ;牛磺酸能明显减少缺血心肌线粒体MDA的生成 ,降低心肌Bax蛋白的表达 ,增加Bc1 2蛋白的表达。结论 :结扎大鼠冠状动脉左前降支导致的心肌缺血损伤与Bc1
Objective:TO investigate the alteration of bcl-2 and bax genes expression in the ischemic myocardium and the influence of taurine. Methods:Myocardial ischemia model of rats was induced by ligation of the anterior branch of the left coronary for 4 h. All animals were divided into three groups: Sham-operation group, Occlusion group, and taurine group (200 mg·kg -1). The level of myocardial mitochondria malondialdehyde (MDA)?Superoxide dismutase(SOD)and Ca 2+-ATPase activeties were determined. The myocardial expression of Bcl-2 and Bax proteins was detected by immunohistochemistry. Results: Coronary occlusion of the rats led to significant increase of MDA content and decreases of SOD?Ca 2+-ATPase activities in the myocardial mitochondria. The expression of Bax protein in myocardial was increased as coronary occlusion,but had no significance change in Bcl-2 protein expression. Taurine reduced the level of MDA and Bax protein in ischemia myocardial and increased myocardial Bcl-2 protein expression?SOD activity and Ca 2+-ATPase activity. Conclusion:The myocardial ischemic injury induced by ligation of the anterior branch of left coronary artery may have relationship with expression of Bcl-2 and Bax proteins.
