摘要
目的 :探讨三氧化二砷 (As2 O3)抑制神经母细胞瘤 SK- N- SH细胞增殖及诱导凋亡的作用。方法 :1采用 MTT法测定不同浓度的 As2 O3在不同时间对 SK- N - SH细胞增殖的抑制率 ;2用 TU NEL - POD法检测不同浓度的 As2 O3诱导 SK- N- SH细胞凋亡的情况。结果 :1各浓度组 As2 O3均可抑制 SK- N- SH细胞增殖 ,而且延长作用时间比增加作用浓度更能增强药物的抑制作用 ;2两个浓度的 As2 O3均可诱导 SK- N - SH细胞凋亡 ,且凋亡指数与浓度高低有关。结论 :As2 O3可以通过诱导神经母细胞瘤 SK- N- SH细胞凋亡而抑制其增殖 。
Objective:To study the role of arsenic trioxide(As 2O 3) inhibiting cell proliferation and inducing apoptosis in Neuroblastoma SK N SH cell line. Methods:①The effect of As 2O 3 inhibiting cell proliferation was assessed by MTT assays. ②Using TUNEL POD method to detect the apoptosis index(AI)of As 2O 3 in SK N SH cell line. Result: ①All concentration groups can inhibite the proliferation of SK N SH cell,and extending act time can improve the effect more than that by way of increasing drug dosage.②All concentration groups can induce apoptosis ,and AI was higher in the high concentration group. Conclution: As 2O 3 can inhibite cell proliferation and induce apoptosis in Neuroblastoma SK N SH cell line. It may be a new promising drug to treat Neuroblastoma.
