摘要
目的 观察牛磺酸对外源性羟自由基诱导心肌细胞凋亡的影响。方法 采用培养的第 2代心肌细胞 ,随机分成 4组 :①正常对照组 :仅用DMEM培养 ;②羟自由基组 :OH-终浓度为 0 1mmol·L-1;③牛磺酸组 :牛磺酸终浓度为 30mmol·L-1;④牛磺酸 +羟自由基组 :牛磺酸 30mmol·L-1+OH-0 1mmol·L-1。观察心肌细胞存活率和形态学 ,流式细胞仪双标法检测细胞凋亡率。结果 ①羟自由基组心肌细胞存活率降低 ,和对照组比较有显著性差异 (P <0 0 5 ) ;牛磺酸 +羟自由基组细胞存活率较羟自由基组高 (P<0 0 5 )。②AnnexinV+ /PI-细胞即凋亡细胞在羟自由基组有较高的发生率 ,明显高于其他各组 (P <0 0 5 ) ;牛磺酸 +羟自由基组的细胞凋亡率显著低于羟自由基组 (P <0 0 5 )。③羟自由基组凋亡心肌细胞呈特征性超微结构。结论 牛磺酸能减轻外源性羟自由基诱导的心肌细胞凋亡。
Objectives To obserev effects of taurine on apoptosis in neonatal rat cardiomyocytes induced by exogenous hydroxyl free radical. Methods ①The second passage of cultured neonatal rat cardiomyocytes were divided randomly into four groups and were co-cultured with hydroxyl free radical (0.1 mmol·L -1 ) and taurine (30 mmol·L -1 ) in serum-free medium for six hours and 24 hours respectively. The survival ratio and morphology of cardiomyocytes and the percentage of Annexin V +/PI - cells by flow cytometry were evaluated. Results ①The survival ratio of cardiomyocytes significantly decreased with 0.1 mmol·L -1 hydroxyl free radical ( P<0.05 vs control) and significantly increased with taurine (30 mmol·L -1 ). ②The percentage of Annexin V +/PI - cells with flow cytometry increased significantly with 0.1 mmol·L -1 hydroxyl free radical and decreased with taurine ( P<0.05 ).Conclusions Taurine attenuates apoptosis of neonatal rat cardiomyocytes induced by exogenous hydroxyl free radical.
