基于荧光通用引物竞争性PCR分析候选基因的表达

摘要 目的:建立用于小家鼠X染色体性发育相关候选基因筛选的一种基于荧光通用引物竞争性聚合酶链式反应(Polymerase Chain Reaction,PCR)定量技术。方法:在小家鼠X染色体上与性发育启动相关的数量性状基因座(quantitative traitlocus,QTL)区段内选择10个候选基因,应用竞争性PCR技术,建立基于荧光通用引物竞争性PCR方案。结果10个候选基因在近交系品系C57BL/6J(B6)和C3H/HeJ(C3)的下丘脑中无表达量差异。结论:本研究,通过实时荧光定量PCR(Real-time quantitative PCR,RT-PCR)技术验证竞争性PCR方案的检测结果准确、可靠。明确10个候选基因与近交系品系B6和C3的性发育启动差异无关。
