摘要
目的建立乳腺癌MDA-MB-231细胞株三维培养模型,观察三维培养条件下乳腺癌细胞的形态、增殖以及HOTAIR表达水平的变化。方法 Matrigel三维培养乳腺癌细胞,采用倒置显微镜观察乳腺癌细胞的生长情况,通过检测DNA含量测定细胞增殖情况,Real time-RCR检测HOTAIR表达水平。结果与二维细胞培养条件相比,Matrigel三维培养的乳腺癌细胞成团生长,细胞数量显著增加,并且HOTAIR mRNA表达水平上调。结论 Matrigel三维培养体系能较好维持乳腺癌MDA-MB-231细胞形态,长链非编码RNA-HOTAIR表达在三维培养时显著升高。
Objective To establish Matrigel three-dimensional(3 D) culture model of breast cancer cells to study the cellular morphology and proliferation and the expression of HOX transcript antisense RNA(HOTAIR). Methods The morphology,distribution and viability of breast cancer cells in three dimensional culture were identified by fluorescence microscopy, the content of DNA was measured, real time-PCR was used to assess the expression of HOTAIR. Results The cells formed multicellular colonies and the number of the cells increased obviously, the expression level of HOTAIR was upregulated in Matrigel 3 D culture than in 2 D culture. Conclusion Matrigel 3 D culture promoted the proliferation of breast cancer cells and upregulated HOTAIR expression.
