摘要
目的检测ZNF711在宫颈癌和宫颈鳞状上皮内病变组织中的表达及定位,分析ZNF711与ER、c-myc蛋白的表达相关性,探讨在宫颈癌中ZNF711的表达及其临床意义。方法应用免疫组织化学染色方法检测ZNF711在108例宫颈癌、宫颈鳞状上皮内病变及宫颈正常组织中的表达及定位,同时应用Western blot检测各组织中的蛋白表达,进一步应用SPSS23.0统计分析ZNF711与ER、c-myc蛋白表达的相关性。结果 ZNF711在宫颈癌、宫颈鳞状上皮内和正常宫颈组织中定位于细胞核;在宫颈鳞状上皮及宫颈癌组织中,随着病变的进展,ZNF711的表达增高。ZNF711的表达与ER的表达呈显著负相关(P<0.01),但与患者年龄、肿瘤分化级别、肿瘤直径大小、浸润深度、临床分期及淋巴结转移及与c-myc的表达无显著相关性。结论 ZNF711参与调控ER的表达,可作为宫颈癌靶向治疗的潜在靶点。
Objective To detect the expression and localization of ZNF711 in cervical cancer and cervical squamous intraepithelial neoplasia, to further understand the correlation between the expression of ZNF711 and the expression of ER and c-myc, and to explore the clinical significance of the expression of ZNF711 in cervical cancer.Methods The expression of ZNF711 was detected using immunohistochemistry and Western blot in 108 cases of cervical cancer, cervical squamous intraepithelial neoplasia and normal cervical tissues. The correlation between the expression of ZNF711 and the expression of ER and c-myc was statistically analyzed using SPSS 23.0. Results The expression of ZNF711 was localized in the nucleus of cervical cancer, cervical squamous intraepithelial neoplasia and normal cervical tissues, and highly expressed in cervical squamous intraepithelial neoplasia and cervical cancer tissues, with negative correlation with the expression of ER(P<0.01), but with no significant correlation with age of patients, differentiation grades and diameter size of tumor, infiltration depth, clinical stage and lymph node metastasis, and no correlation with the expression of c-myc. Conclusion ZNF711 is involved in regulating the expression of ER, and might be a potential target for targeted therapy of cervical cancer.
