摘要
目的观察槲皮素对胶质瘤C6细胞增殖的影响及其对脑胶质瘤大鼠脑组织DR5表达的影响,探讨槲皮素对脑胶质瘤C6细胞增殖的调控作用。方法将体外培养的脑胶质瘤C6细胞按照槲皮素浓度分成25、50、75及100μmo l/L 5个处理组和对照组(二甲亚砜),采用MTT比色法检测槲皮素对胶质瘤C6细胞增殖的影响,Western blotting法检测100μmo l/L处理24h后的细胞和对照组细胞死亡因子受体5(DR5)的蛋白表达。选取雌性Wistar大鼠30只,随机分为3组:假手术组、模型组和槲皮素治疗组,每组10只。建立大鼠C6胶质瘤模型,于接种后17d处死各组大鼠,采用免疫组织化学法和Western bloting法检测肿瘤组织DR5蛋白表达水平的变化。结果槲皮素药物浓度增加或作用时间的延长,均能显著增强对C6细胞的增殖抑制作用。100μmo l/L槲皮素处理24h后,C6细胞DR5表达显著升高,P<0.01。在体研究时发现,槲皮素能够上调脑胶质瘤大鼠脑组织DR5蛋白表达的平均光密度值,P<0.01。结论槲皮素对C6细胞增殖抑制作用具有浓度、时间依赖性,上调DR5表达可能是其作用机制之一。
Objective To study the effect of quercetin on the proliferation of C6 cells and the expression of death receptor 5(DR5) in brain tissue of glioma rats, and to explore the regulatory effect and mechanism of quercetin on the proliferation of glioma cells. Methods C6 glioma cells cultured in vitro were divided into 5 groups according to the concentration of quercetin: 25, 50, 75 and 100μmol/L and control group(dimethyl sulfoxide). MTT was used to observe the effects of quercetin on the proliferation of C6 glioma cells. The expression of DR5 in C6 glioma cells was detected by Western blot. A total of 30 female rats were randomly divided into 3 group(n=10 in each group:sham group, model group and quercetin treatment group.The rat model of C6 glioma was established,and animals were killed at 17 day after tumor graft. The expression of DR5 was observed by immunohistochemical method and Western blot. Results Quercetin significantly enhanced the inhibition of C6 cell proliferation in a concentration or time manner. The expression level of DR5 in C6 cells was increased significantly after treatment with 100μmo l/L quercetin for 24 hours(P<0.01). Quercetin upregulated the mean optical density of DR5 protein expression in brain tissue of glioma rats(P<0.01).Conclusion Quercetin inhibits the proliferation of C6 cells in a concentration-and time-dependent manner, which is related to the upregulation of DR5 expression.
