摘要
目的观察伤害性感觉神经元过表达Relish、Dorsal及Kir2.1后,果蝇三龄幼虫对伤害性刺激的敏感度及逃避行为的变化,探讨NF-κB基因家族对机体伤害性逃避反应的影响。方法应用基因遗传学方法,分别在果蝇伤害性感觉元内特异性过表达Relish、Dorsal及Kir2.1,观察果蝇幼虫对紫外线照射产生反应的潜伏期、逃离时间及逃避行为。结果过表达Relish、Dorsal及Kir2.1组的潜伏期和逃离时间均比对照组延长,卷曲次数无明显变化,滚动次数及滚动百分率低于野生型对照组。结论伤害性感觉神经元过表达Relish或Dorsal可导致果蝇幼虫出现与抑制神经元兴奋性相似的行为反应,即会出现伤害性逃避反应的延迟。
Objective To observe the changes of sensitivity and escape behavior of Drosophilar third instar larvae to noxious stimuli-ultraviolet light ray after overexpression of Relish, Dorsal and Kir2.1. to explore the effects of NF-κB family proteins on depression-like behavior in organisms. Methods Using genetic method, expression of Relish,Dorsal and Kir2.1 specific in nociceptors of Drosophila. 40 larvae in the same size of each group were selected, and each larva was observed under ultraviolet irradiation. The incubation period, escape time, percentage of rolling and average number of rolling of ultraviolet light exposure are compared to wild type. Results Under ultraviolet light irradiation, the incubation period and escape time of over-expressed Relish, Dorsal and Kir2.1 groups were longer than those of control group;the percentage of rolling and average number of rolling of larvae over-expressing Relish, Dorsal and Kir2.1 was lower than that of the wild-type control group. Conclusion Overexpression of Relish or Dorsal in nociceptive neurons caused Drosophila larvae to exhibit a behavioral response same as suppress neuronal excitability, a delay behavior in response to noxious escape.
