The effect of chromosome structure maintenance protein(SMC1) on cell multipolarity

摘要 目的研究染色体结构维持蛋白SMC1不同表达水平对细胞多极形成的影响。方法分别采用分子生物学和慢病毒包装的方法建立稳定过表达SMC1和稳定沉默SMC1 (shSMC1)-HCT116细胞系,而后用Western Blot的方法检测稳定细胞株的构建是否成功,最后应用免疫荧光方法检测过表达与低表达SMC1两种情况对细胞多极分裂的影响。结果 Western blot及免疫荧光结果显示HCT116细胞在过表达SMC1与低表达SMC1两种情况与正常HCT116细胞相比均显著增加了细胞多极的比例且有统计学意义(P<0.05)。结论 SMC1与细胞的多极行为密切相关,对基因组稳定性的维持发挥了重要作用。 Objective To study the effect of different expression levels of chromosome structure maintenance protein(SMC1)on cell multipolar formation.Methods Overexpressed and silencing SMC1(shSMC1)-HCT116 stable cell line are established respectively by molecular biology and lentiviral packaging.The effect of overexpression and low expression of SMC1 on cell multipolar division was studied by immunofluorescence.Results Western blot and immunofluorescence showed that overexpression of SMC1 and lowexpression of SMC1 significantly increased the multipolar ratio of cells compared with normal HCT116 cells.Conclusion SMC1 is closely related to the multipolar formation of cells and plays an important role in the maintenance of genomic stability.
